56% of female university students get drunk in record time, Spanish study suggests

Dec. 28, 2012 ? Researchers from the HealthyFit group at the University of Vigo have studied university students' lifestyles; their analysis, which includes alcohol and illegal drug consumption habits, sport and food, concludes that most students indulge in unhealthy behaviour. One of the main results of the study points to the high consumption of alcohol.

"The amount drunk per unit of time is higher among women. In other words, even though male students drink more often, females do so more intensively in shorter periods of time, which is known as binge drinking," explained Jos? M? Cancela Carral, co-author of the study published by the Journal of Environmental Research and Public Health.

Researchers randomly selected 985 students from different degree courses and in different years at the University of Vigo.

Of the females interviewed, 51.2% lead a sedentary lifestyle, while the percentage in males is 41.7%. When analysing students who maintain an appropriate level of physical activity, 38.6% of males do physical exercise, as opposed to only 20.9% of women.

"We were also surprised by the high consumption of illegal drugs among university students -- 44.9% of men and 30.9% of women -- which we understand could lead to significant future health problems, mainly related to the nervous system," underlined the researcher.

Anomalous attitudes to food were more evident among women (16.6%), although also present among men (8.8%). "However, the statistical analysis showed that this parameter depended on the degree the student was studying for," added Cancela. Such attitudes were much more common among those studying degrees related to education (19.2%) than among those studying courses related to health (6.3%).

Health network

Spanish universities set up a Healthy University Network in 2008, a project for healthy living for universities from all over Spain, the Spanish University Rectors' Association, the Ministry of Health, Social Services and Equality, the Ministry of Education, Culture and Sport and some regional public health entities.

In the researcher's opinion, at many universities this network was nothing more than "a simple first step to get on the list and nothing else"; hence transversal content should be implemented in study plans related to food, physical exercise and healthy habits.

"In the light of the results, training and information courses are required in these areas, together with healthy leisure -- not just sports facilities -- to set up university guidance services for a healthy student lifestyle," concludes Cancela.

Genetic predisposition

A recent study published in the PNAS journal argues that the gene called RASGRF2 could be related to a predisposition to getting drunk.

According to scientists, this gene regulates the predisposition to drink excessive amounts of alcohol as it influences mesolimbic dopamine neuron activity, which is one of the ways dopamine is taken from one region of the brain to another.

Gunter Schumann, one of the main authors of the study, explained that even though we should not consider said gene to be the main cause -- as there are many environmental factors and other genes involved -- the study thereof helps to explain why some people are more vulnerable to alcohol than others.

Works and Days ? An Anatomy of a Most Peculiar Institution

A Campus-full of Contradictions

Almost everything about the modern university is a paradox. It has become a sort of industry gone rogue that embraces practices that a Wal-Mart or Halliburton would never get away with. It is exempt from scrutiny in the fashion that the Left ceased talking about renditions or Guantanamo Bay once Barack Obama was elected, or a Code Pink goes after a NRA official in the way it would never disrupt a hearing on Fast and Furious. In other words, the university is one of the great foundations of the Left, and so is immune from the sort of criticism that otherwise is daily leveled against other institutions.

So let?s take a 10-minute stroll through the campus and learn why costs soar even as students are ever more poorly educated.

The Curriculum

A student?s life on campus is a zero-sum game. For each elective like ?The modern comic book,? or ?Chicana feminisms? or ?Queering the text,? students have no time (or desire to) take more difficult and instructive classes on the British Enlightenment or A History of World War I or Classical English Grammar. (Yes, despite the relativist, anti-hierarchical university, concepts really do exist ?like ?more instructive.?) The former are mostly therapeutic classes, entirely deductive, in which the point is not to explore an intellectual topic by presenting the relevant facts and outlining the major controversies, while sharpening students? inductive reasoning and empirical objectivity, as well as improving their English prose style and mastering grammar and syntax in their written work.

The result is perhaps a fourth of the liberal arts courses ? many would judge more like 50% ? would never have been allowed in the curriculum just 40 years ago. They tend to foster the two most regrettable traits in a young mind ? ignorance of the uninformed combined with the arrogance of the zealot. All too often students in these courses become revved up over a particular writ ? solar power, gay marriage, the war on women, multiculturalism ? without the skills to present their views logically and persuasively in response to criticism. Heat, not light, is the objective of these classes.

Why are these courses, then, taught?

For a variety of practical reasons: 1) often the professors are rehashing their doctoral theses or narrow journal articles and are not capable of mastering a wider subject (e.g., teaching a class in ?The Other in Advertising? ?is a lot easier than a systematic history of California); 2) the quality of today?s students is so questionable that the social sciences have stepped up to service the under-qualified, in the sense of providing courses, grades, and graduation possibilities; 3) the university does not see itself as a disinterested nexus of ideas, where for a brief four years students are trained how to think, given a corpus of fact-based knowledge about their nation and world, and expected to develop an aesthetic sense of art, music, and literature. Instead college is intended as a sort of boot camp for the progressive army, where recruits are trained and do not question their commissars.

So the new curriculum in the social sciences and humanities fills a need of sorts, and the result is that today?s graduating English major probably cannot name six Shakespearean plays; the history major cannot distinguish Verdun from Shiloh; the philosophy major has not read Aristotle?s Poetics or Plato?s Laws; and the political science major knows very little of Machiavelli or Tocqueville ? but all of the above do know that the planet is heating up due to capitalist greed, the history of the United States is largely a story of oppression, the UN and the EU offer a superior paradigm to the U.S. Constitution, and there are some scary gun-owning, carbon-fuel burning, heterosexual-marrying nuts outside the campus.

If we ask why vocational and tech schools sprout up around the traditional university campus, it is because they are upfront about their nuts-and-bolts, get-a-job education: no need to worry about ?liberal arts? or ?the humanities? ? especially given that the universities? General Education core is not very general and not very educational any more. Yes, I am worried that the University of Phoenix graduate has not read Dante, but more worried that the CSU Fresno graduate has not either, and the former is far more intellectually honest about that lapse than the latter.

Note here the illiberal nature of allowing highly paid faculty to indulge their curricular fantasies at the expense of indebted students who pay a great deal for a great deal of nothing. Is there a provost or a dean in America that can say to faculty, ?That is not a real course, and so won?t be taught at our real university?? Does the shop foreman let the welder choose his own project?

Auto sales look good for '13, but for the 'cliff'

It?s been a disappointing holiday season for many?retailers, but U.S. car dealers have been busily ringing up sales?as the year wraps up.

If preliminary numbers for December hold up, 2012 could prove to be the industry?s best year since the long recession and painfully weak recovery began. And 2013 is?shaping up to be even better ? unless auto sales fall off the fiscal cliff. ?

?I clearly hope we get some bipartisan effort to avoid the fiscal cliff,? said Ford Motor Co. Chairman Bill Ford Jr. ?It?s vitally important for the economy that we work this out.?

That's true for the auto industry, in particular. After nearly half a decade of struggles, carmakers are ending 2012 on a positive note. November saw demand for new cars, trucks and crossovers surge at a double-digit pace. In fact, a number of makers including Audi, BMW, Honda, Nissan and Porsche are setting all-time sales records for the month. By the end of November, Hyundai had already beaten its previous full-year record set in 2011.

December, meanwhile, is expected to see another gain of around 15 percent, year-over-year, according to a forecast by LMC Automotive. The consulting firm has now upped its final forecast for all of 2012 to 14.5 million and conservatively anticipates another comfortable jump next year.

?The U.S. light-vehicle sales market continues to be a bright spot in the tremulous global environment,? said Jeff Schuster, LMC?s senior vice president of forecasting.? ?The only major roadblock ahead for the U.S. market is the fiscal cliff. Assuming that hurdle is cleared, 2013 is one step closer to a stable and sustainable growth rate for autos, with volume above the 15 million unit mark.?

Barring that setback, analysts anticipate a steady shift in the sort of vehicles Americans have been buying.?

The upcoming North American International Auto Show will feature an assortment of new products, and the debut of the all-new 2014 Chevrolet Corvette and Chevy?s next-generation Silverado pickup are likely to be two of the star attractions. But the floor of Detroit?s Cobo Center will be covered with a variety of downsized models, including Honda?s pint-sized Urban SUV Concept and the new Lincoln MKC compact luxury crossover.

Ford's?B- and C-segment models ? subcompacts like the updated 2013 Fiesta and compacts???have jumped from 13 percent?of the overall U.S. market to 24 percent since 2004, according to Mark Fields,?Ford?s chief operating officer.?And the trend is expected to continue at an even faster pace going forward.

If anything, Americans are downsizing powertrains at an even more rapid rate, trading in V-6s for inline-fours and even abandoning traditional V-8s for high-tech turbo V-6s. Six-cylinder engines now account for more than half of the demand in Ford?s big F-Series pickups, Fields said.

While demand for smaller models is soaring, sales of some traditional American vehicles have remained strong ? pickups in particular. Credit the strong upturn in the housing market in recent months, said Mark Reuss, president of North American operations at General Motors.?

While the so-called ?personal use? segment has largely dried up, there?s significant pent-up demand among contractors and other professional for pickups and other full-size trucks, something Detroit?s Big Three hope to use to their advantage as they continue to hold off import competitors like Nissan and Toyota.

Pent-up demand is likely to float all boats, says analyst Joe Phillippi, from the largest to the smallest market segments. It helps, he adds, that automotive interest rates are at or near historic lows.

On the downside, makers have been cutting back incentives as demand outstrips supply, especially of more popular models like the Hyundai Elantra and the new Honda Accord. There are still bargains to be had on slower-selling models, but research by TrueCar.com reveals that incentives have slipped to their lowest levels in several years even as transaction prices ? the price customers actually pay ? have risen to near-record levels.

And that is likely to translate into record industry earnings, at least in North America. Ironically, the market is running counter to trends in much of the rest of the world. European car sales have been in freefall all year, and without a clear resolution of the Continent?s economic crisis, few see any short-term solution.? Even China, which has seen annual growth rates approaching 100 percent over the past decade, has started to stumble.

So, makers from around the world are once again looking to the U.S. to carry the load. The good news is that ?the forecast ahead looks even better,? Toyota?s top U.S. executive, Jim Lentz, recently said, adding that ?analysts expect we will reach 16 million in just a few short years.?

That?s still well short of the U.S. all-time record of 17.5 million vehicles sold in 2005, but it?s still a welcome turnaround from the disastrous slump the industry has only just left behind.

Daily Kos: My Grandmother has brain cancer

Her name is Margaret. She married my grandfather decades ago and they had a baby. His name was John Thomas Schehr. He was my father. Shortly after her husband died. He was a sailor in the Navy. He died of spinal meningitis when my father was a wee babe.

Margaret remarried to a man named Joseph LaGreca, and my father took his name. Joseph LaGreca and Margret than had another child whom they also named Joseph.

But Joesph the elder was a lout. He inflicted unimaginable abuse upon my father and my uncle, and it stayed with them for the course of their lives. I can't imagine what my Grandmother went through, but she stayed, she honored their vow and she endured. How she did so is beyond my reckoning.

My father left home at a young age and joined the navy as his father before had done, but he stayed a devoted son and he never spoke against his grandfather in my presence. Each year we would get in the car and visit Grandma and Grandpa for Thanksgiving, Christmas and Easter, my brother and sister with my mother and often the family dog would all make the trip upstate to Ulster County New York, and though our mother had told us of what our Step-Grandfather had put our father through my dad never once spoke of it, and he always dutifully made the trip up to his parents, to his mother and step father. Though my father honored his step-father I can only imagine the love he had for his mother, my grandmother, in staying a truly devoted son all those years.

And when my father died we grew distant. That was years and years ago, but though we stayed in touch by phone it was rare when I saw my grandmother.

And now she is almost gone.

The doctors say she has a tumor pressing up against her brain. She no longer responds to people, and only days ago she had troubling recognizing my sister when she had gone to visit. She has developed pneumonia and the fever is getting worse. The doctors have asked if we would sign a "Do Not Resuscitate" waiver, or something of that sort.

And then it all comes home. This is what we are fighting about. All politics is personal. It's not just about the ideas or the principles, it is about actual people and how to make their lives better. After all my advocacy for health care reform this is when it hits home, because we love people and we want them to be safe and cared for, and because sometimes those people are real people we know and love and sometimes we will never meet them at all, but goddamit we can do better for people, better than this, and that is worth fighting for. I see the nurse caring for my Grandmother and I ask myself what can we do for her to make her life better, I think of that nurse's family and I wonder if they struggle and I ask myself what can we do to make their lives better, more fulfilling, more prosperous, so that this short struggle we all endure on this silly spinning orb can be made slightly better, if only for a time. I saw my grandmother yesterday for perhaps the last time, and though she was never awake I told her I loved her and I wished her peace with my father at last. I believe she heard me.

Strange behavior: New study exposes living cells to synthetic protein

Dec. 27, 2012 ? One approach to understanding components in living organisms is to attempt to create them artificially, using principles of chemistry, engineering and genetics. A suite of powerful techniques -- collectively referred to as synthetic biology -- have been used to produce self-replicating molecules, artificial pathways in living systems and organisms bearing synthetic genomes.

In a new twist, John Chaput, a researcher at Arizona State University's Biodesign Institute and colleagues at the Department of Pharmacology, Midwestern University, Glendale, AZ have fabricated an artificial protein in the laboratory and examined the surprising ways living cells respond to it.

"If you take a protein that was created in a test tube and put it inside a cell, does it still function," Chaput asks. "Does the cell recognize it? Does the cell just chew it up and spit it out?" This unexplored area represents a new domain for synthetic biology and may ultimately lead to the development of novel therapeutic agents.

The research results, reported in the advanced online edition of the journal ACS Chemical Biology, describe a peculiar set of adaptations exhibited by Escherichia coli bacterial cells exposed to a synthetic protein, dubbed DX. Inside the cell, DX proteins bind with molecules of ATP, the energy source required by all biological entities.

"ATP is the energy currency of life," Chaput says. The phosphodiester bonds of ATP contain the energy necessary to drive reactions in living systems, giving up their stored energy when these bonds are chemically cleaved. The depletion of available intracellular ATP by DX binding disrupts normal metabolic activity in the cells, preventing them from dividing, (though they continue to grow).

After exposure to DX, the normally spherical E. coli bacteria develop into elongated filaments. Within the filamentous bacteria, dense intracellular lipid structures act to partition the cell at regular intervals along its length. These unusual structures, which the authors call endoliposomes, are an unprecedented phenomenon in such cells.

"Somewhere along the line of this filamentation, other processes begin to happen that we haven't fully understood at the genetic level, but we can see the results phenotypically," Chaput says. "These dense lipid structures are forming at very regular regions along the filamented cell and it looks like it could be a defense mechanism, allowing the cell to compartmentalize itself." This peculiar adaptation has never been observed in bacterial cells and appears unique for a single-celled organism.

Producing a synthetic protein like DX, which can mimic the elaborate folding characteristics of naturally occurring proteins and bind with a key metabolite like ATP is no easy task. As Chaput explains, a clever strategy known as mRNA display was used to produce, fine-tune and amplify synthetic proteins capable of binding ATP with high affinity and specificity, much as a naturally occurring ATP-binding protein would.

First, large libraries of random sequence peptides are formed from the four nucleic acids making up DNA, with each strand measuring around 80 nucleotides in length. These sequences are then transcribed into RNA with the help of an enzyme -- RNA polymerase. If a natural ribosome is then introduced, it attaches to the strand and reads the random sequence RNA as though it was a naturally-occurring RNA, generating a synthetic protein as it migrates along the strand. In this way, synthetic proteins based on random RNA sequences can be generated.

Exposing the batch of synthetic proteins to the target molecule and extracting those that bind can then select for ATP-binding proteins. But as Chaput explains, there's a problem: "The big question is how do you recover that genetic information? You can't reverse transcribe a protein back into DNA. You can't PCR amplify a protein. So we have to do all these molecular biology tricks."

The main trick involves an earlier step in the process. A molecular linker is chemically attached to the RNA templates, such that each RNA strand forms a bond with its newly translated protein. The mRNA-protein hybrids are exposed to selection targets (like ATP) over consecutive rounds of increasing stringency. After each round of selection, those library members that remain bound to the target are reverse-transcribed into cDNA (using their conveniently attached RNA messages), and then PCR amplified.

In the current study, E. coli cells exposed to DX transitioned into a filamentous form, which can occur naturally when such cells are subject to conditions of stress. The cells display low metabolic activity and limited cell division, presumably owing to their ATP-starved condition.

The study also examined the ability of E. coli to recover following DX exposure. The cells were found to enter a quiescent state known as viable but non-culturable (VBNC), meaning that they survived ATP sequestration and returned to their non-filamentous state after 48 hours, but lost their reproductive capacity. Further, this condition was difficult to reverse and seems to involve a fundamental reprogramming of the cell.

In an additional response to DX, the filamentous cells form previously undocumented structures, which the authors refer to as endoliposomes. These dense lipid concentrations, spanning the full width of the filamented E. coli, segment the cells into distinct compartments, giving the cells a stringbean-like appearance under the microscope.

The authors speculate that this adaptation may be an effort to maintain homeostasis in regions of the filamentous cell, which have essentially been walled off from the intrusion of ATP-depleting DX. They liken endoliposomes to the series of water-tight compartments found in submarines which are used to isolate damaged sections of the ship and speculate that DX-exposed cells are partitioning their genetic information into regions where it can be safely quarantined. Such self-compartmentalization is known to occur in some eukaryotic cells, but has not been previously observed in prokaryotes like E. coli.

The research indicates that there is still a great deal to learn about bacterial behavior and the repertoire of responses available when such cells encounter novel situations, such as an unfamiliar, synthetic protein. The study also notes that many infectious agents rely on a dormant state, (similar to the VBNC condition observed in the DX-exposed E. coli), to elude detection by antibiotics. A better understanding of the mechanisms driving this behavior could provide a new approach to targeting such pathogens.

The relative safety of E. coli as a model organism for study may provide a fruitful tool for more in-depth investigation of VBNC states in pathogenic organisms. Further, given ATP's central importance for living organisms, its suppression may provide another avenue for combating disease. One example would be an engineered bacteriophage capable of delivering DX genes to pathogenic organisms.

Mandela close to medical care at home

JOHANNESBURG (AP) ? The doctors treating former South African leader Nelson Mandela believe he should remain in Johannesburg for now to be close to medical facilities that can provide care to the 94-year-old, the government said Thursday.

Mandela left a hospital Wednesday evening after nearly three weeks of treatment there, and was brought to his home in the Johannesburg neighborhood of Houghton. The anti-apartheid icon, also known by his clan name, Madiba, has spent more time in recent years in the rural village of Qunu in Eastern Cape province, where he grew up.

Mandela's grandson, Mandla Mandela, said he hopes "it won't be too long before he's with us back in Qunu, where he belongs," but acknowledged that the doctors' assessment is critical to any decision to travel.

"It can be a strenuous trip," the grandson said in an interview with eNCA, a South African television news channel. "We will await the feedback from the doctors as to when he will be fit and ready to come back home."

Mandela was admitted Dec. 8 to a hospital in the South African capital of Pretoria, 50 kilometers (30 miles) north of Johannesburg. The ex-president was treated for a lung infection and also had a procedure to remove gallstones.

"Where Madiba goes, in which period, in which times, is a matter that is entirely dependent on his own wishes. Whatever he wishes, we will do," presidential spokesman Mac Maharaj told eNCA.

"But right now, the doctors have considered it necessary and good that he should be in Houghton so that he's close to all the facilities where we can give him high care," Maharaj said.

Maharaj noted that Mandela had been in good spirits while receiving President Jacob Zuma and other visitors while he was hospitalized.

"Madiba was doing well, but as you know, when you're recovering there are ups and downs, slight ups and downs, and the doctors are looking for a steady progress and that began to be registered over the last few days," the spokesman said.

Mandela is revered around the world as a symbol of sacrifice and reconciliation, his legacy forged in the fight against apartheid, the system of white minority rule that imprisoned him for 27 years. The Nobel laureate served one five-year term as president after South Africa's first democratic elections in 1994.

Maharaj said Mandela is able to see his family "more easily" now that he is at home, but he appealed to the public to respect the former president's privacy "to allow the best possible conditions for his full recovery."

Afghan policewoman kills US adviser, police say

KABUL, Afghanistan (AP) ? An Afghan policewoman shot and killed an American adviser outside the police headquarters in Kabul on Monday, a senior Afghan official said. The circumstances of the killing were not immediately clear but the shooting could be another insider attack by Afghans against their foreign allies.

A NATO command spokesman, U.S. Air Force Lt. Col. Lester T. Carroll, said the shooter was taken into Afghan custody shortly after the incident. The slain adviser was a contractor but the name and nationality of the deceased were being withheld, Carroll said.

Deputy Police Chief Mohammad Daoud Amin said it has not been determined whether the American adviser's death was intentional or accidental. He said an investigation was under way and declined to give more details of the incident, which occurred in central Kabul on the compound housing the governor of the city and near a number of key ministries.

"We can confirm that a civilian police adviser was shot and killed this morning by a suspected member of the Afghan uniform. The suspected shooter is in Afghan custody," Carroll said. He said Afghan and NATO officials were still trying to confirm initial reports that the shooter was a policewoman.

The killing came just hours after an Afghan policeman shot five of his colleagues at a checkpoint in northern Afghanistan late Monday. The attacker then stole his colleague's weapons and fled to join the Taliban, said deputy provincial governor in Jawzjan province, Faqir Mohammad Jawzjani.

More than 60 international allies, including troops and civilian advisers, have been killed by Afghan soldiers or police this year, and a number of other insider attacks as they are known are still under investigations. NATO forces, due to mostly withdraw from the country by 2014, have speeded up efforts to train and advise Afghan military and police units before the pullout.

The surge in insider attacks is throwing doubt on the capability of the Afghan security forces to take over from international troops and has further undermined public support for the 11-year war in NATO countries.

In February, two U.S. soldiers -- Lt. Col. John D. Loftis and Maj. Robert J. Marchanti, died from wounds received during an attack by an Afghan policeman at the Interior Ministry in Kabul. The incident forced NATO to temporarily pull out their advisers from a number of ministries and police units and revise procedures in dealing with Afghan counterparts.

More than 50 Afghan members of the government's security forces also have died this year in attacks by their own colleagues. Taliban militants claim such attacks reflect a growing popular opposition to both foreign military presence and the Kabul government.

In Sunday's attack, Jawzjani, the provincial official, said the attacker was an Afghan policeman manning a checkpoint in Dirzab District who turned his weapon on five colleagues before fleeing to the militant Islamist group.

This is a recipe my father used back in the 40?s. Many years ago, I found it among his things, but never made it until tonight. Now I will never make any other version. It?s really airy yet creamy, with just the right balance of booze.

I Twitter/Facebooked about this tonight, and am posting it here after several requests, after all, we still have New Year?s to go through!

Note that this recipe uses raw eggs. If this scares you, I doubt you?d be on this site anyway, but normal warnings apply. Use good, clean eggs and break them correctly (not on the side of a bowl).

Egg Nog

Beat 8 egg whites until stiff peaks.

Add 1/2 cup sugar and beat until incorporated. Set aside for a moment.

Beat 8 egg yolks and 1/2 cup sugar.

Put the whites back on the mixer, and fold in the beaten yolks

At a slower speed, add 1 qt heavy cream, 1 pt whole milk, beat some more.

Add 2 cups rum and 1/2 cup bourbon.

Take 20 mg Lipitor.

Pour into glasses, dust with freshly ground nutmeg

Autistic-like behaviors can be partially remedied by normalizing excessive levels of protein synthesis in the brain, a team of researchers has found in a study of laboratory mice. The findings, which appear in the latest issue of Nature, provide a pathway to the creation of pharmaceuticals aimed at treating autism spectrum disorders (ASD) that are associated with diminished social interaction skills, impaired communication ability, and repetitive behaviors.

"The creation of a drug to address ASD will be difficult, but these findings offer a potential route to get there," said Eric Klann, a professor at NYU's Center for Neural Science and the study's senior author. "We have not only confirmed a common link for several such disorders, but also have raised the exciting possibility that the behavioral afflictions of those individuals with ASD can be addressed."

The study's other co-authors included researchers from the University of California, San Francisco (UCSF) and three French institutions: Aix-Marseille Universite'; Institut National de la Sant et de la Recherche Mdicale (INSERM); and Le Centre National de la Recherche Scientifique (CNRS).

The researchers focused on the EIF4E gene, whose mutation is associated with autism. The mutation causing autism was proposed to increase levels of the eIF4E, the protein product of EIF4E, and lead to exaggerated protein synthesis. Excessive eIF4E signaling and exaggerated protein synthesis also may play a role in a range of neurological disorders, including fragile X syndrome (FXS).

In their experiments, the researchers examined mice with increased levels of eIF4E. They found that these mice had exaggerated levels of protein synthesis in the brain and exhibited behaviors similar to those found in autistic individualsrepetitive behaviors, such as repeatedly burying marbles, diminished social interaction (the study monitored interactions with other mice), and behavioral inflexibility (the afflicted mice were unable to navigate mazes that had been slightly altered from ones they had previously solved). The researchers also found altered communication between neurons in brain regions linked to the abnormal behaviors.

To remedy to these autistic-like behaviors, the researchers then tested a drug, 4EGI-1, which diminishes protein synthesis induced by the increased levels of eIF4E. Through this drug, they hypothesized that they could return the afflicted mice's protein production to normal levels, and, with it, reverse autistic-like behaviors.

The subsequent experiments confirmed their hypotheses. The mice were less likely to engage in repetitive behaviors, more likely to interact with other mice, and were successful in navigating mazes that differed from those they previously solved, thereby showing enhanced behavioral flexibility. Additional investigation revealed that these changes were likely due to a reduction in protein productionthe levels of newly synthesized proteins in the brains of these mice were similar to those of normal mice.

"These findings highlight an invaluable mouse model for autism in which many drugs that target eIF4E can be tested," added co-author Davide Ruggero, an associate professor at UCSF's School of Medicine and Department of Urology. "These include novel compounds that we are developing to target eIF4E hyperactivation in cancer that may also be potentially therapeutic for autistic patients."
